จากกรุงเทพมหานคร ใช้เวลาขับรถประมาณ 2 ชั่วโมงไปยังท่าเรือบ้านเพ จังหวัดระยอง ซึ่งจะมีเรือเฟอร์รี่ หรือ speed boat ข้ามฟากอยู่ตลอดทุกครึ่งชั่วโมงหรือทุกชั่วโมงโดยปกติอยู่แล้ว ที่จะพาข้ามฟากไปที่ท่าเรือหน้าด่านบนเกาะเสม็ด แต่อยากจะจะแนะนำว่าให้ออกจากรุงเทพแต่เช้า เพื่อให้มาถึงท่าเรือเร็วที่สุด จะได้มีเวลาเที่ยวเกาะเสม็ด 1 วันเต็มๆ อย่างไรก็ตามใครที่ขับรถยนต์ส่วนตัวมา บริเวณรอบ ๆ ท่าเรือบ้านแพมีที่ฝากจอดรถหลายแห่ง ราคาประมาณวันละ 100 บาท สามารถสอบถามเจ้าหน้าที่แถวนั้นได้เลย เมื่อเรียบร้อยแล้วก็ซื้อตั๋วเรือ ซึ่งใช้เวลานั่งเรือประมาณ 40 นาทีก็จะมาถึงเกาะเสม็ด
